I know Mail for Exchange only available for Nokia N- and E- series, but Nokia 6121 is also a S60 device, so will MfE work? I know there's alternative like RoadSync.

No, I tried and it reports "installation failed" part way through. Specs of OS and hardware are exactly the same as the E and N series so it isn't a matter of it can't, it's just they lock the option out to make you buy E or N series.

I'm using RoadSync but it isn't as good as MailForExchange (for me anyway) and also the 6121 doesn't allow the inbox to be installed to the standby screen. This means you have to rely on hearing the tone to know you have new email and then navigate to the RoadSync mailbox manually.

(I'm waiting patiently for the flood of 900MHz UMTS E series announcements next week at CTIA Wireless ;-) )

Ok, cheers. Guess I will wait for next ultimate phone. N82 vs E51 vs Iphone 2 vs (anything S60+UMTS900mhz) LOL... As long as they support Exchange ActiveSync.
