I haven't had much experience with USB flash drives and a couple day ago it was my first time using Ready Boost on a 4gb USB Flash drive. I had to remove the Flash drive because I had to temporarily use the slot for a boot-able disk. I put the USB back in, I thought I could Format it and try Ready boost again, but it said that there was a program running in it and I couldn't Format it. Does this mean my USB is still actively using Ready boost or my USB is messed up?

You will have to right-click, Properties and stop using ReadyBoost on that USB key.

Do you find it actually makes any difference. When I used it, it didn't seem to do anything to performance at all.

You can only have one. And getting more memory is a lot more effective.

Correct, with Windows 7 you can have up to eight devices actually.

ReadyBoost was an interesting stop gap measure but consumer USB flash disks are actually pretty slow and get slower over time (it seems) then ram and SSD's got cheaper, so they are far far better options now.

Eg: You can get an Intel 330 SSD 120GB for ~$140 now.

and it seems some USB sticks just aren't usable at all I have two 4GB sticks and 1 16GB stick which just refuse to run ready boost

I wouldn't bother with Ready boost, it was really for when Vista first came out and most PCs had like 512mb or 1GB of RAM. RAM is so cheap now days, just buy some more
